Design Notes

1)Choice of tag sequence.The length of tag can influence the folding or assembly of GmPilA monomers.So we had to choose a glue peptide pair that is short enough.SnoopTag and SnoopCatcher is a suitable pair. 2)Choice of position to merge SnoopTag:GmPilA monomers need to fold and assemble properly and a merged peptide may influence this process.After reading relevant articles, we found that the C-terminus of GmPilA monomer is projected out from the pilus surface and previous researches found that a short peptide merged to its C-terminus would not significantly interfere with this process.
